I think there was one of General LeMay's raids over Osaka that involved more than 500 B-29's if you can imagine that sight. I would have loved to have seen that.
There were other single raids of 200, 300, and 400 B-29's', but I'm pretty sure that the Osaka raid was the biggest.

Do you mean the "Peacemaker", marchplumber?
It looked a lot like the B-29, but it was "next-gernation" and bigger (check out the comparison photo in this link here; second photo down):
